The Dakshin Trisagar LRM 1200 starts from Ginger, Thiruvananthapuram (0 km) and heads south to Sunset Viewpoint, Kanyakumari (100 km), followed by Copper Leaf Hotels, Tirunelveli (184.5 km) and St. Mary's Boys Higher Secondary School, Thoothukudi (228 km).

The route then continues to Shree Priyam Residency, Ramanathapuram (355 km) before reaching Arichal Munai Beach, Dhanushkodi (430 km), the easternmost point of the route.

Riders then retrace the route back through Ramanathapuram (505 km) and continue towards Madurai (631 km), Dindigul (695.5 km) and Palani (748.5 km).

From Palani, the route heads west to Karpagam University, Coimbatore (837.5 km) and Varsha Lodge, Alathur, Palakkad (900 km), before continuing through Mannuthy, Thrissur (937 km), Muvattupuzha (1012.5 km), Kottayam (1069 km) and Adoor (1126 km).

The final leg takes riders back to Ginger, Thiruvananthapuram (1208 km), completing the Dakshin Trisagar LRM.
